Objectives: To evaluate the contribution of genetic and environmental factors for stability in, comorbidity with, and life events related to personality disorders.
Methods: DSM-IV criteria for 10 personality disorders were assessed using structured interviews in 2801 Norwegian twins in early adulthood (age 28; range 19-36) and again in 2282 twins 10 years later. Biometric models were used on criteria counts to analyze the genetic and environmental factors for PDs and: 10-year stability, comorbidity with common mental disorders and psychotic-like experiences, substance abuse, and stressful life events.
Results: Heritability of PDs were estimated to be 40% to 20% heritable. The 10-year stability estimated as tetrachoric correlation of PDs ranged from .35 to .58. Genetic variants influencing PDs were estimated to be the same over 10 years, the environmental factors were estimated to change 74-84% over the same period. Genetic and environmental factors for comorbidity between PDs and common mental disorders have different dimensionality and structure. The link between PDs and substance use are dependent on the specific criteria endorsed rather than at the disorder level. Stressful life events are important for the development of PDs and substance use over and beyond what can be explained by life-course selection due to genetic risk.
Conclusion: Although PDs at specific assessments are mostly influenced by environmental factors, genetic factors are more important in explaining stability and comorbidity. The unequal structure of genetic and environmental factors leads to a mismatch between the observed comorbidity between PDs and mental disorders and the expected commonality in genetic factors. Aggregation of symptoms and disorders according to the estimated genetic structure could improve the precision of molecular genetic studies. This longitudinal twin study has also shown that PDs are related to stressful life events, mental disorders, and substance abuse over and beyond what can be explained by genetic risk.

Genomics of borderline personality disorder.

Factors such as early trauma or abuse are well-established environmental risk factors for the development of Borderline Personality Disorder (BPD). Compared to other mental disorders, little attention has been paid to the contribution of genetic risk factors. The present symposium aims to give an overview of how family and twin studies, and molecular genetic studies can contribute to the understanding of the disorder, and its relationship to risk factors and comorbid disorders:
Eivind Ystrom will present on how twin studies can be used to investigate genetic and environmental risk factors contributing to comorbidity between BPD, mental disorders, and substance abuse. Using interview data from a population-based twin study, Ystrom and colleagues have found that genetic factors are the main contributors to the 10-year stability of BPD. Genetic factors for BPD are to a great extent overlapping with the risk for antisocial PD and substance use disorders. However, BPD is related to stressful life events, internalizing symptoms and personality disorders, and substance abuse over and beyond what can be explained by common genetic risk factors.
Ralf Kuja-Halkola will present how national health register data can be used to investigate genetic contributions. Using twins and siblings, familial aggregation of clinical BPD diagnosis was investigated, and the heritability of BPD was estimated to 46%. The familial co-aggregation between BPD and attention deficit hyperactivity disorder (ADHD) was estimated, e.g. full siblings of individuals with ADHD diagnosis have a 2.8 times increased risk of having a BPD diagnosis compared to full siblings of individuals without ADHD diagnosis. Additionally, the co-aggregation in full siblings between BPD and a range of psychiatric and somatic disorders, and behavioral traits will be presented.
Fabian Streit will present the initial results of the largest genome-wide association study (GWAS) which systematically investigates the contribution of common genetic variants to BPD. In a meta-analysis including over 6000 cases, and 500,000 controls. The results highlight first associated genes, also related to comorbid disorders, such as post-traumatic stress disorder (PTSD), or ADHD. Polygenic risk scores predicted up to 3% of the variance in case-control status. Positive genetic correlations were observed with other psychiatric disorders, most pronounced with PTSD, depression, ADHD, and anxiety. Additional analyses indicated genetic correlations with factors such as somatic disorders, pain, mood swings, childhood maltreatment, and material deprivation.
Guy Hindley will present molecular genetic investigations of mood instability, a key component of BPD. A recent GWAS identified a strong positive genetic correlation of the trait with depression but weak correlations with bipolar disorder and schizophrenia. However, genetic correlation does not capture shared genetic variants with mixed effect directions. By applying statistical tools which capture mixed effect directions, the extensive genetic overlap between mood instability and all three mental disorders could be demonstrated, irrespective of their genetic correlation. This overlap was used to identify 246 novel genetic loci associated with mood instability. Biological characterization highlighted a convergence on synaptic gene-sets. These findings indicate shared neurobiological mechanisms between mood instability and diverse mental disorders.

The genomics of psychiatric symptom differentiation in early life.

Background: An emerging body of evidence suggests that genetic risk for psychiatric conditions is diffuse, probabilistic in nature, and overlaps substantially across different domains. However, psychiatric conditions are diagnosed and treated as specific disorders based on differentiated symptom clusters. In the context of generalised genetic risk, it remains to be determined how distinct psychiatric conditions emerge.
The prospects of using genomic data to predict individual-level risk for psychiatric conditions are at present unclear. For prediction to be useful, it needs to be both sensitive (identifying who is at risk) and specific (at risk for what). While increasing sample sizes for genetic discovery may boost sensitivity, the generalised nature of genetic risk implies that identifying predictors of differentiation of psychiatric symptoms may be needed to increase specificity. The Norwegian Mother, Father, and Child Cohort Study (MoBa) is a large population-based pregnancy cohort with genotype and phenotype data, ideally suited to address these questions.
Methods: We used the Child Behavior Checklist to assess behavioural and emotional difficulties at ages 1.5, 3, and 5. Symptom¬ differentiation was operationalised as the relative level of these domains over time (behavioural difficulties – emotional difficulties = differentiation). This means that individuals with high scores have relatively more behavioural than emotional difficulties, and the inverse for low scores. These scores can be compared to overall difficulties (behavioural + emotional = total). Following a preregistered analysis plan, we estimated latent growth trajectories of differentiation across early childhood, as well as mediation effects of differentiation on symptoms of psychiatric conditions at age 8. We tested associations between 10 polygenic scores (PGS) for core neurodevelopmental and psychiatric conditions and the extent (intercept) and rate (slope) of the differentiation process. The analytic sample comprised 57,387 genotyped children (including 12,347 pairs of full siblings) from MoBa. We used multilevel structural equation models to account for unmeasured familial confounding, and corrected for multiple comparisons using FDR.
Results: ADHD PGS predicted the extent and rate of differentiation toward behavioural difficulties in early childhood (βintercept = 0.10 [0.09, 0.11]; βslope = 0.02 [0.02, 0.03]), and the extent of differentiation at age 5 in turn mediated 33-37% of the PGS associations with ADHD symptoms at age 8. The ADHD PGS association was smaller for total difficulties (βintercept = 0.05 [0.04, 0.06]; βslope = 0.01 [0.01, 0.02]). Schizophrenia PGS predicted differentiation toward emotional difficulties at age 5 (βintercept = -0.03 [-0.04, -0.02]; βslope = -0.01 [-0.02, -0.01]), and bipolar disorder PGS predicted differentiation toward behavioural difficulties (βintercept = 0.02 [0.01, 0.03]; βslope = 0.01 [0.01, 0.02]) – though in both cases effect sizes were substantially smaller.
Discussion: We identify genomic predictors of symptom differentiation in early life. Results support a direct effect of ADHD PGS on differentiation toward behavioural difficulties, independent of individuals’ total burden of symptoms. PGS for schizophrenia and bipolar disorder differentially associates with the development of emotional and behavioural difficulties in early childhood. This research may help refine risk predictions in child psychiatry, an important step to enable early prevention.

Modest overlap in the genetic influences underlying normal and abnormal personality.
Show summary
Background and aims: Substantial correlations between normal and abnormal personality traits have been found, but there is still little knowledge regarding the extent to which the underlying genetic and environmental influences are shared.
Methods: 2,282 Norwegian twins were assessed for DSM-IV criteria for personality disorders using structured interviews. Ten years later years the twins were re-interviewed, and normal personality was assessed using the Big Five Inventory. Multivariate twin models were fitted to the five domains of normal personality and dimensional measures of each of the personality disorders individually.
Results: While the heritability of the number endorsed criteria for the personality disorders ranged from 0.2 to 0.41, the proportion of genetic variance that was unique to each PD ranged from 19% for Avoidant to 79% for Schizotypal, with a median of 58% across all PDs. Conversely, the proportion of environmental variance unique to the personality disorder criteria ranged from 79% to 99%, with median of 97%.
Conclusion: Our results suggest that a considerable proportion of the genetic variance and virtually all environmental variance is unique to the personality disorders, and not shared with the five broad domains of normal personality.

Avoidant and obsessive-compulsive personality disorder traits from early to middle adulthood.

Backgroundand Aims: The stability over time for avoidant personality disorder (AVPD) and obsessive-compulsive personality disorder (OCPD) has previously been found to be moderate. However, the mechanisms underlying this temporal development are not well understood. In the present study we estimate the influence of genetic and environmental factors on stability and change in AVPD and OCPD.
Methods: AVPD and OCPD traits were assessed using the Structured Interview for DSM-IV Personality in 2,793 young adult twins from the Norwegian Institute of Public Health Twin Panel (wave 1). On average 10 years later (wave 2) 2,282 of these were re-interviewed. Longitudinal biometric models were fitted to dimensional representations of AVPD and OCPD.
Results: The number of endorsed sub-threshold criteria for both PDs decreased 31% from wave 1 to wave 2. The correlations between the numbers of traits endorsed at the two time points were 0.54 for AVPD and 0.37 for OCPD. Genetic factors contributed 67% and 53%, respectively, to the stability of AVPD and OCPD. The correlations between the genetic factors influencing AVPD and OCPD at the two time points were 1.00 and 0.72 respectively, while the environmental influences correlated 0.26 and 0.23, respectively. The correlation between the stable risk factors for AVPD and OCPD was 0.39 of which 63% was attributable to genetic influences.
Conclusion: Both AVPD and OCPD were moderately stable from young to middle adulthood. Stability was in large part due to genetic factors, whereas environmental influences contributed to change.

Borderline and antisocial personality disorder traits in early to middle adulthood: A longitudinal twin study.
Show summary
Background and Aims: Antisocial personality disorder (ASPD) and borderline personality disorder (BPD) share genetic and environmental risk factors. Little is known about the temporal stability of these etiological factors in adulthood.
Methods: DSM-IV criteria for ASPD and BPD were assessed using structured interviews in 2,282 Norwegian twins in early adulthood and approximately 10 years later. Longitudinal biometric models were used to analyze the number of endorsed criteria.
Results: The mean criterion count for ASPD and BPD decreased 40% and 28% respectively from early to middle adulthood. Rank-order stability was 0.58 for ASPD and 0.45 for BPD. The best-fitting longitudinal twin model included only genetic and individual-specific environmental factors. Genetic effects, both those shared by ASPD and BPD, and those specific to each disorder remained completely stable. The unique environmental effects, however, changed substantially, with a correlation across time of 0.19 for the shared effects, and 0.39 and 0.15 respectively for those specific to ASPD and BPD. Genetic effects accounted for 71% and 72% of the stability over time for ASPD and BPD respectively. The genetic and environmental correlations between ASPD and BPD were 0.73, and 0.43 respectively at both time points.
Conclusion: ASPD and BPD traits were moderately stable from early to middle adulthood, mostly due to genetic risk factors which did not change over the10 year assessment period. Environmental risk factors were mostly transient, and appear to be the main source of phenotypic change. Genetic liability factors were, to a large extent, shared by ASPD and BPD.

Breastfeeding and personality traits: the impact of maternal negative affectivity and self-efficacy on breastfeeding. The Norwegian Mother and Child Cohort Study.

Objective: To measure the impact of the personality traits of Negative Affectivity and General Self-Efficacy measured during pregnancy to predict breastfeeding status at 4-months postpartum, adjusting for cesarean section, premature birth, parity, maternal age, and years of maternal education.
Design: Prospective cohort study.
Setting: The data collection was conducted as part of the Norwegian Mother and Child Cohort Study at the Norwegian Institute of Public Health. The national sample of 29,581 mothers completed assessment of Negative Affectivity and General Self-Efficacy at gestation weeks 17 and 30 and completed a self-report questionnaire at 6-months postpartum capturing detailed information about nutrition from birth thru 6 months.
Outcome Measures: Mothers were classified with a cut-off at 4 months in the groups full breastfeeding, mixed breastfeeding, and bottle feeding.
Results: We found that each interval of high Negative Affectivity increased the odds ratio of mixed breastfeeding (OR, 1.39; 95% CI, 1.27-1.51) and bottle feeding (OR, 1.56; 95% CI, 1.39-1.76), compared to full breastfeeding. Each interval of high General Self-Efficacy decreased the odds ratio of bottle feeding at 4 months (OR, 0.89; 95% CI, 0.83-0.96) but not of mixed breastfeeding (OR, 1.00; 95% CI, 0.95-1.04), compared to full breastfeeding.
Conclusions: Our results support the hypothesis that Negative Affectivity and General Self-Efficacy are important antenatal predictors of breastfeeding 4 months postpartum. Adequately addressing the concerns and difficulties of mothers high in Negative Affectivity and low in General Self-Efficacy during prenatal and antenatal care could help prevent difficulties to sustain full breastfeeding.

Svekkelse av kognitive funksjoner som hukommelse og språkforståelse er en utbredt følge av traumatisk hjerneskade. De senere årene har flere studier undersøkt feilfri læring ("errorless learning") som metode for kognitiv rehabilitering etter traumatisk hjerneskade. Studiene rapporterer at feilfri læring gir bedre læringsresultater enn læring gjennom prøving og feiling
("errorful learning"). Feilfri læring baseres på et prinsipp
for læring der anledningen til å gjøre feil reduseres eller elimineres. Den aktuelle pasientgruppen regnes som spesielt sårbar for at de feil som gjøres i en læringssituasjon, kan forstyrre innlæringen av riktig informasjon. Så langt har forskning på dette feltet vært konsentrert om kontrollerte, eksperimentelle situasjoner, og har i liten grad dokument klinisk anvendelse av feilfri læring som metode for rehabilitering. Målsetningene for denne studien er 1) å undersøke mulige forbedringer eller raffineringer av læringsteknikker basert på feilfri læring, og 2) evaluere feilfri læring i klinisk rehabilitering av personer som har vært utsatt for traumatisk hjerneskade.

Bakgrunn: Angst og depresjon hos mødre påvirker flere prosesser hos barn som er viktig for utvikling av prososial atferd. Det finnes ikke undersøkelser som har sett på sammenhengen mellom prososial atferd hos barn i førskolealder og mødre med angst og depresjon.
Mål: Å undersøke i en ikke-klinisk populasjon om det var en sammenheng mellom prososial atferd hos 3 år gamle barn og mødrenes angst og depresjon når barnet var 6, 18 og 36 måneder. Det andre målet var å undersøke om effekten av mors angst og depresjon ble
moderert av om barnet vokste opp sammen med andre barn i husholdningen og om barnet var i barnehage eller lignende på dagtid. Det siste målet var å undersøke om det var en sammenheng mellom varigheten på angsten og depresjonen hos mødrene og barnas prososiale atferd.
Metode: Datainnsamlingen ble utført som en del av Den norske mor og barn undersøkelsen(MoBa) ved Nasjonalt folkehelseinstitutt. Utvalget besto av 5314 mødre og barn. Mødrene besvarte spørreskjemaer når de var i 18. svangerskapsuke og da barna var 6, 18 og 36 måneder gamle. Mødrene rapporterte om barnas prososiale atferd, da barna var 3 år gamle. Dette ble målt med prososial skala i SDQ. Mødrene rapporterte om egen angst og depresjon som målt med SCL-8 da barna var 6, 18 og 36 måneder gamle. Prososial atferd ble inndelt i normalskårer (80. prosentil) og lav skårer (20. prosentil).
Resultater: Lav prososial atferd hos 3 år gamle barn ble predikert av angst og depresjon hos mor da barnet var 6, 18 og 36 måneder, også etter at det var kontrollert for sosiodemografiske variabler og faktorer i barnets proksimale miljø. Både det å være hjemme med mor eller far i 18. levemåned og hjemme med mor i tredje leveår, predikerte større sannsynlighet for å ha lav prososial atferd, sammenlignet med om barnet var i barnehage eller lignende. Effekten av
mors angst og depresjon på barnets prososialitet ble ikke moderert av om barnet var i barnehage eller lignende. Barnas lave prososiale atferd ble predikert av tilbakevendende angst og depresjon hos mor i barnas tre første leveår, men ikke av vedvarende eller kortvarig angst
og depresjon.
Konklusjon: Angst og depresjon hos mødre har en betydning for 3 år gamle barns utvikling av prososial atferd. Mors angst og depresjon påvirker barnets utvikling av prososial atferd fra det er spedbarn og til det er 3 år gammelt. Selv ikke å tilbringe tid i barnehage, park, hosdagmamma, praktikant eller med far, reduserer denne risikoen.

Denne studiens hovedmål var å undersøke hvilken rolle personlighetstrekk etter
femfaktormodellen, og manipuleringstaktikker (Buss, 1992), spilte for tilfredsheten i et
parforhold. Det ble postulert at manipuleringstaktikker medierer sammenhengen mellom
partners personlighet og egen tilfredshet. Dette ble gjort ved å undersøke følgende
sammenhenger: a) forholdet mellom tilfredshet og personlighet, b) forholdet mellom
manipuleringstaktikker og personlighet, c) forholdet mellom manipuleringstaktikker og
tilfredshet, d) forholdet mellom personlighetstrekk og tilfredshet, mediert av
manipuleringstaktikker. Studien har også undersøkt kjønnsforskjeller og personlighetslikhet
mellom ektefeller. Denne studien har vært et selvstendig forskningsprosjekt, hvor det har blitt
oversatt og satt sammen et spørreskjema bestående av selvrapportert personlighet (BFI),
tilfredshet i parforhold (RAS), samt selvrapportert og partnerrapportert bruk av
manipuleringstaktikker (ToM) i parforhold. Det ble samlet inn data fra 242 par med barn i
grunnskolen, hvor hver av ektefellene svarte selvstendig på hvert sitt skjema. Dataanalysene
inkluderte regresjonsanalyse og strukturlikningsmodellering.
Resultatene viste at nevrotisisme hadde en negativ sammenheng med tilfredshet, mens
omgjengelighet og samvittighetsfullhet hadde en positiv sammenheng med tilfredshet. Videre
at egne personlighetstrekk, heller enn ektefellens personlighetstrekk alene var av størst
betydning for egen tilfredshet. Det var mange sammenhenger mellom personlighetsrekk og
manipuleringstaktikker for både kvinner og menn. De fleste fantes mellom omgjengelighet,
nevrotisisme og samvittighetsfullhet i forhold til egenrapporterte manipuleringstaktikker.
Bruken av taktikker generelt og umodenhetstaktikker spesielt viste negativ sammenheng med
tilfredshet for både kvinner og menn. En strukturell likningsmodell på tilfredshet og
personlighet mediert av taktikker viste at egen tilfredshet var predikert av egne
personlighetstrekk, og ikke predikert av ektefellens personlighetstrekk alene, men mediert av
partners manipuleringstaktikker. Resultatene tyder på at egen personlighet har mer å si for
egen tilfredshet enn ektefelles personlighet. Det ser ut til at nevrotisisme og mangel på
omgjengelighet og samvittighetsfullhet, sammen med umodenhetstaktikker gir et solid
amalgam som forringer ektefellens tilfredshet.
